Gossaert was born around 1478 in Maubeuge, Northern France. His nickname as a painter, Mabuse, derives from the Netherlandish appellation of the town.

In 1503 he became a member of the painter’s guild in Antwerp and in 1508–1509 he travelled to Rome with Philip of Burgundy, then ambassador of Margaret of Austria to Pope Julius II. Influenced by the art he saw in Rome, Gossaert became an influential supporter of the representation of biblical and mythological subjects with naked figures. His style must have appeared shockingly new and avant-garde to his Northern European patrons, and he can be regarded as one of the first Renaissance painters in Northern Europe.
